Sample Requirements

A. Minimum sample collection and storage requirements for CHILDREN:

  1. Infant (<1 year): 3 mL EDTA (purple top) anticoagulated whole blood for Class I, II
  2. Child (1 year-10 years): 5 mL EDTA for Class I, II
  3. Child (>10 years): 10 mL EDTA for Class I, II
  4. Contact the HLA Laboratory when the WBC count is < 5.0 X 103 for required sample amount.
  5. Samples may be refrigerated (4°C) or stored at room temperature.

B. Sample collection and storage requirements for ADULTS:

  1. HLA Class I (A,B,C,) or Class II (DR,DQ, DP) Typing
    1. Specimen: Collect 20 mL EDTA, mix thoroughly. When Class I and Class II are requested 20 mL EDTA will be sufficient.
    2.    
      Collect 40 - 60 mL EDTA when the WBC count
      is < 5.0 X 103. Contact the HLA laboratoy for assistance in determining required sample amount.
    3. Storage temperature: Refrigerated (4°C) or room temperature.


  2. B-27 Typing
    1. Specimen: Collect 5 mL EDTA, mix thoroughly.
    2. Storage temperature: Refrigerated (4°C) or room temperature.
      Note: ACD may be substituted for EDTA anticoagulated samples for DNA molecular testing.

  3. Antibody Screen
    1. Specimen: Collect 10 mL whole blood in a clot tube (red top).
    2. Storage temperature: Store and transport at 4°C. If not shipped within 48 hours of collection, separate and freeze serum. Send on ice.

  4. HLA Crossmatch
    1. Recipient Specimen: Collect 10 mL whole blood in a clot tube (red top).
    2. Recipient Specimen Storage Temperature: Store and transport at 4°C. If not sent with in 48 hours of collection, separate and freeze the serum. Send on ice.
    3. Donor Specimen: Collect 30 mL ACD (yellow top) anticoagulated whole blood.
    4. Donor Specimen Storage Temperature: Store and transport at room temperature.

Blood Systems, Inc. has two Donor Testing Service (DTS) laboratories. Our second laboratory, in Bedford, Texas was opened in 1999. Both DTS laboratories are Food and Drug Administration (FDA) licensed facilities. Both DTS laboratories also hold current certifications for the Clinical Laboratory Improvement Act (CLIA) as well as accreditation with the American Association of Blood Banks and the New York State Department of Health.
